MGF IGF-Ec

MGF (mechano growth factor) is the name given to a splice variant of the IGF-1 gene, IGF-1Ec in humans (the rodent homolog is IGF-1Eb), produced locally in skeletal muscle in response to mechanical loading or injury rather than circulating systemically like liver-derived IGF-1Ea. Unlike mature IGF-1, the C-terminal E-domain peptide cleaved from the IGF-1Ec precursor does not bind the IGF-1 receptor with the same affinity and is therefore studied as a distinct signaling entity through alternative, less-characterized pathways. The E-domain peptide (24 amino acids in humans) has been proposed to activate satellite cell proliferation independently of the IGF-1 receptor.

Riddoch-Contreras and colleagues demonstrated in SOD1(G93A) mouse models of ALS that a synthetic MGF C-terminal peptide rescued motoneurons and improved muscle function — a neuromuscular application distinct from MGF's originally proposed role in satellite-cell activation for muscle hypertrophy research. Subsequent work from multiple groups has described IGF-IEc expression across cardiac, hepatic, neural, and testicular tissues, expanding the scope of potential E-domain peptide research beyond skeletal muscle.
